Abstract:A fragment containing HPV16 E6-E7 ORFs with its autologous Long Control Region (HPV16 LCR E6-E7) was inserted into eukaryotic expression vector pSV2neo in sense orientation. We successfully transformed NIH3T3 cells in vitro with the constructed plasmid. The transformed cells showed anchorage-independent growth and can cause tumor in nude mice. Southern blot analysis of DNAs from the transformed cells and tumor tissues in nude mice showed integration of the constructed plasmid into the host DNA. The results indicated that HPY16 E6-E7 transforming genes might play a critical role in the development of human cervical carcinoma.
