稻瘟净(Blasticidin S)脱氨酶基因的克隆

摘    要:从一株抗稻瘟净(BS)的Aspergillus terreus菌中克隆到一个blastieidinS脱氨酶基因,命名为bsrAS。DNA序列分析表明bsrAS不含内含子。编码区长390bp,编码130个氨基酸。将bsrAS转化到稻瘟菌中,能使受体菌表达出BS脱氨酶的活性,从而产生抗药性。该基因可作为抗药标记基因使用,建立稻瘟菌的基因转化系统。

Abstract:A structural gene of blasticidin S (abbreviated:BS) deaminase has been aloned from a Aspergillus terreus strain that is resistance to blasticidin S,a fungicide used for the control of rice blast.The gene was named bsrAS and its restriction map has been constructed in this study.The nucleotide sequence shows that it contains no intron.The coding region contains 390 bp and codes for a putative protein of 130 amino acids,The gene was introduced into Pyricularia oryzae by transformation and it conferred BS resistance to all transformants.It was demonstrated that the BS deaminase was expressed and the regulatory seguence of the gene was functional in P.oryzae.DNA hybridization data confirmed that the gene was integrated into the genome of recipient cells. This achievement results in a new drug resistance selective marker for transformation of P.oryzae.
