铕螯合剂DTTAEuNa标记流行性出血热单克隆抗体的实验研究

用Ｎ′（对异硫氰基苄基）二乙三胺Ｎ１，Ｎ２，Ｎ３四乙酸铕钠（ＤＴＴＡＥｕＮａ）作为螯合剂，标记不同蛋白浓度的流行性出血热单克隆抗体（ＥＨＦＭｃＡｂ），经ＳｅｐｈａｋｅｘＧ５０凝胶柱分离标记的单抗分子，通过对层析液的紫外吸收峰处的蛋白光密度，时间分辨荧光强度以及免疫活性测定表明：二批Ｅｕ标记单抗的比活性分别为７８和１４７个Ｅｕ３＋／ＥＨＦＭｃＡｂ，标记回收率分别为４１％和４２％，标记物的免疫活性良好，可用于流行性出血热的抗原及抗体检测

Experimental study of europium labeled monoclonal antibody against epedemic hemarrhagic fever virus

To develop a more sensitive method of detectiong epidemic hemarrhagic fever virus (EHFV) antigen and antibody, the effect of N' (p isothiocyanatobenzy1) diethylenetriamine N -1 ,N -2 ,N -3 , tetraacetate europium sodium(DTTAEuNa)Labeled minoclonal antibody against epidemic hemarrhagic fever virus(EHFV McAb) was reserched.DTTAEuNa reacts on the protein of EHFV McAb.The Eu 3 labeled EHFV McAb was separated from escess reagent by gel filtration on a column of Sephadex G 50.The flusorescence and ultraviolet absorption for the fractions were monitored and the Eu 3 EHFV McAb immunoactivity was detected.The specific activity for two group Eu 3 EHFV McAb was 7.8Eu 3 and 14.7Eu 3 /EHFV McAb molecule and the recovery rate of Eu 3 EHFV McAb was 41% and 42% respectively.The Eu 3 EHFV McAb immunoactivity was excellent.The EHFV McAb labeled by DTTAEuNa is able to detect the EHFV antigen and antibody.
