紫鸭跖草花粉母细胞减数分裂细胞核和染色体含有F—肌动蛋白

摘    要:肌动蛋白间接免疫荧光标记实验观察到紫鸭跖草花粉母细胞减数分裂前期Ⅰ细胞核,中期┘染色体,二分体和四分体均发现较明亮的黄绿色荧光,说明其中含有肌动蛋白。经TRIT-C鬼等环肽荧光标记后,减数分裂前Ⅰ细胞核,二分枝及中期Ⅰ染色体均发现较明亮的红色荧光;CD处理后细胞核和染色体的荧光明显减弱或没有荧光,说明其中存在F-肌动蛋白。

Abstract:Pollen mother cells(PMCs) of Tradescantia virginiana were labelled with anti-actin antibody and FITC-conjugated secondery antibody. Prowinent yellow-green fluorescenceReceived May 5, 1998, revision received september 1,1998suppolted by the Trans-Century Ttaining Program Foundation for the Talents by the State Education Commissionof Chinawas found in meiohc prophase I nuclei, metaphase I chromosomes, dyads and tetrads,indicating the presence of actin in them. Observations upon TRITC-conjugatedphalloidin-labelled specimens showed that the meiotic prophase I nucleus, metaphase Ichromosomes and dyads in PMCs without special treatment all sent forth distinctivesred fluorescence, but the fluorescence of the nuclei and chromosomes was very weakor even lost in the PMCs exposed to cytochalasin D before fixation, demonstratingthat F-actin is present in meiotic nuclei and chromosomes. When double labelling ofanti-actin antibody and phalloidin was applied, the same meiohc nuclei andchromosomes were found to emanate yellow-green fluorescence representing actin atthe excitation wavelength of HTC, and red fluorescence representing F-actin atexcitation wavelength of TRITC, respectively. The FITC-type fluorescence andTRITC-type fluorescence shared the same distribution among the meiotic nuclei andchromosomes.
