A mutant in Escherichia coli energy-uncoupled for lactose transporta defect in the lactose-operon

Abstract:From a K12 strain (X71) a mutant (X71-54) has been isolated which possesses a severe defect in the active transport of thiomethyl-galactoside without reduction in total membrane carriers as assayed by o-nitrophenylgalactoside entry. Several lines of evidence suggest that the mutant is defective in the energy-coupling mechanism to the lactose transport system. This energy-uncoupled property and the lactose operon were co-transduced (via P1) into a lactose deleted strain (Ca 77).
