Abstract:Carrot suspension cells were culturedby single cell technique and found thatgrowth and the content of pigment,espe-cially,the ability of embryogenesis ofsingle cell clones were great different.Under the light and on the basic MS me-dium where carrot callus could undergoembryogenesis,about half of clones ex-pressed differentiation,however;amongthem,half had a normal morphology,14% abnormal,19% roots only,18%embryos only.But,in the dark,majori-ty of clones could not express differen-tiation,34% could.Among them,46%formed roots,6% had a normal morpho-logy,28% abnormal.Callus derived fromsingle cell clones could not differentiateand could not still express differentiationon the medium of different combinationof auxin,cytokinin,gibberellic acid andabscisic acid.It's suggested that theylost morphogical ability in heridity. Different staining density and zymo-graphy of isoperoxidase were found incomparison of callus and differentiatedtissue,however;both types of tissue de-rived from single cell clones expressedsimilar difference also.
