摘 要: | 双特异性磷酸酶3(dual-specificity phosphatase 3,DUSP3)是一种双特异性蛋白酪氨酸磷酸酶,其不仅可使磷酸酪氨酸残基去磷酸化,同时也能使磷酸丝/苏氨酸残基去磷酸化。因此,DUSP3具有多样的底物特异性,除了丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)ERK、JNK和p38底物外,还可以作用于其他信号通路的蛋白质,通过MAPK依赖性或非依赖性机制参与细胞周期调控、细胞增殖、衰老、凋亡、迁移以及血管生成、基因组稳定性等多种生物学反应,与肿瘤的发生发展密切相关,是一个潜在的有价值的治疗靶点。本文就DUSP3的生物学特征及其在肿瘤研究中的进展进行了综述。

Abstract: | Dual-specificity phosphatase 3(DUSP3)is a member of the dual-specificity protein tyrosine phosphatases that can dephosphorylate both tyrosine and serine/threonine residues.DUSP3 has various substrates.In addition to MAPKs ERK,JNK and p38,DUSP3 can also target proteins involved in other signaling pathways,playing important roles in cell cycle regulation,proliferation,senescence,apoptosis,migration,angiogenesis,genomic stability through MAPK-dependent or MAPK-independent mechanisms.DUSP3 has been closely implicated in the occurrence and development of tumors.Therefore,it is a potentially valuable target for therapy.This review summarized the biological characteristics of DUSP3 and its advances in tumor research. |
