结直肠癌中Claudinl、ZEB-1和E-cadherin的表达及意义

摘    要:目的探讨Claudinl、ZEB-1和E-cadherin在结直肠癌中的表达及三者之间的关系与临床病理的联系。方法免疫组织化学技术检测51例结直肠癌及癌旁组织中Claudinl、ZEB-1和E—cadherin蛋白的表达情况。结果在结直肠癌组织中Claudinl、ZEB-1、E—cadherin的表达率分别为68.6%、23.5%、56.9%,癌旁组织分别为37.3%、0、98.0%。三种蛋白在结直肠癌组织及癌旁组织中的表达差异均具有显著性(P〈O.01)。Claudinl及ZEB-1的表达率在III_IV期组(87.5%、41.7%)高于I-II期组(51.9%、7.4%;P〈0.01、P〈0.01),有淋巴结转移组(90.9%、45.5%)高于无淋巴结转移组(51.7%、6,9%;P〈0.01、P〈0.01),ZEB-1表达率在高级别组(42.1%)高于低级别组(12.5%;P〈0.05)。E—cadherin的表达率在I—II期组(74.1%)高于Ⅲ—IV期组(37.5%;P〈O.05),无淋巴结转移组(72.4%)高于有淋巴结转移组(36.4%;P〈0.05)。Claudinl、ZEB-1与E-cadherin的表达均呈负相关(P〈0.05),Claudinl与ZEB-1的表达呈正相关(P〈0.05)。结论Claudinl、ZEB-1和E-cadherin在结直肠癌发展,浸润和转移中起着重要作用,三者可能存在协同作用。

Abstract:Objective To study the expression of Claudinl,ZEB-1 and E-cadherin in colorectal cancer, and to investigate the relationship between them and clinicopathologic parameters. Methods Immunohistochemical staining was conducted to detect the expression of Claudinl,ZEB-1 and E-eadherin proteins in 51 samples of colorectal tumor tissues and 51 samples of normal colorectal mucosa. Results The positive rates of Claudinl,ZEB-1 and E-cadherin were 68.6% ,23.5%,56.9% in colorectal tumor and 37.3~//00,0%,98.0% in normal colorectal mucosa, with a significant difference between the two groups (P〈0. 01). The expression rates of Claudinl and ZEB-1 in III-IV stages (87.5%,41.7%) was higher than I-II stage (51.9%, 7.4 % ;P〈0.01,P〈0. 01), as in the cases with lymph node metastasis (90. 9 %, 45.5 %) than in those without metastasis (51.7%,6,9%~P~0.01,P%0.01). The expression rate of ZEB-1 in high grade (42.1%) was also higher than in low grade (12.5% ,P〈0. 05). The expression rate of E-cadherin in I-II stages (74.1%) was higher than in III-IV stages (37.5% ,P%0. 05) ,and higher in the cases with no lymph node metasta- sis (72.4%)than in those with metastasis (36.40% ,P〈0.05). Positive correlations were observed between Claudinl and ZEB-1 in CRC tissues(P〈0.05). The expression of E-cadherin was negatively correlated with that of Claudinl and ZEB-1 (P〈0.05). Conclusion Expressions of Claudinl, ZEB-1 and E-cadherin may be involved in the progression,invasion and metastasis of CRC. Interactions between Claudinl,ZEB-1 and E-cadherin may promote CRC progression.
