摘    要: 在哺乳动物细胞中克隆表达的人生长激素,经等电点和乙醇沉淀后,再用亲和层析法,获得产品。经对该产品的各项生化指标分析鉴定和活力测定,所得结果都与天然人生长激素相符合,证明其性质相同。

Abstract:Biosynthetic hGH by eukaryotic genetic engineering is of great potential value in medical and agricultural application.The purification method of such hGH was established through studies of features of immunoaffinity materials in this paper.The cell culture medium of hGH-gene transformed mouse L cell lines was purified by iso-precipitation at pH 4.90 and one step immunoaffinity chromatography.The highly purified hGH was determined to have identical immunitive and biological activity with native one by radio-immunoassay (RIA) and radioreceptor assay (RRA) respectively.The purified hGH was determined to have molecular weight of 21 500 daltons by SDS-PAGE, isoelectric point of pH 4.90 by isoelectric focus and Phe as N-terminal amino acid by Dansyl Chloride terminal assaying method.All of the cha-raterizations conform with those of the native hGH.
