摘    要:以分子信标为报告分子,核酸适体为识别分子,发展了一种新的凝血酶检测方法.含有分子信标互补序列的核酸适体探针与凝血酶结合后,分子信标的荧光信号下降,从而得到凝血酶的浓度信息.该方法快速、灵敏,核酸适体探针无需荧光标记、设计简单,检测限达到0.83nmol/L.

Abstract:A novel assay for thrombin was developed,in which molecular beacons(MBs) and aptamers were used as reporters and recognizers,respectively.Probes composed of the cDNA sequence of MB and aptamer of thrombin were used.The fluorescence of MB decreased once aptamer bound with thrombin.The fluorescence decrease of MBs was quantitatively dependent on the concentration of thrombin.The detection limit was 0.83 nmol/L.The design of probes was simple and no labeling was required.This assay was rapid and sensitive.It could be used for other proteins by just changing the aptamer.
