菲立磁标记食蟹猴骨髓间充质干细胞的脑内移植示踪研究

摘    要:中枢神经系统损伤后的再生修复问题一直是神经科学领域关注的重点之一,骨髓间充质干细胞移植治疗拓宽了人类中枢神经系统损伤的治疗前景,而非侵入性的磁共振成像能活体追踪移植细胞,评价移植效果。应用菲立磁标记食蟹猴骨髓来源的间充质干细胞,在脑立体定位仪引导下,自体脑内移植。结果显示,菲立磁标记间充质干细胞的有效率高达90%以上,移植区磁共振影像呈明显的低信号改变。标记的间充质干细胞移植后在脑内存活,并向周围的脑实质内迁移。移植8周后,发现移植细胞通过血管向对侧脑部迁移,但并未发现移植细胞向神经细胞分化。这些结果提示,菲立磁可用于标记、追踪脑内移植的食蟹猴骨髓间充质干细胞,标记的移植细胞可在脑内存活、迁移。

Tracking Observation of Cynomolgus Monkey Bone Marrow Mesenchymal Stem Cells Labeled with Feridex after Autologus Transplantation

Abstract:Central nervous system regeneration and repair after injury has been concerning by the researchers in the field of neuroscience.Transplantation of bone marrow-derived mesenchymal stem cells(MSCs) into the injured brain or spinal cord may provide therapeutic benefit.Non-invasive high resolution imaging is required to track the cells and evaluate clinical relevance after cell transplantation.In this study,under sterile condition,cynomolgus monkey monocytes were isolated by density gradient centrifugation following an iliac puncture,and adherent cultured in vitro.Feridex-poly-l-lysine(FE-PLL) complexes were used to magnetically label MSCs.Labeled cells were autologous transplanted into the brain of cynomolgus monkey with the guidance of MRI and stereotaxic devices.The results showed that the efficiency of MSCs labeled with Feridex was up to 90 percentages.After the transplantation,MRI showed remarkable low signal change in the transplantated point.Labeled MSCs could survival in the striatum region,and migrate to the brain tissue around the injection site.8 weeks after transplantation,labeled cells migrated through blood vessels to the contra-lateral brain,but there was no indication of that the transplanted cells differentiated into nervous cells.Our results indicate that Feridex can be used to label and track MSCs in vivo.
