Analysis of adenine nucleotides and metabolic intermediates in mature and senescent leaf tissue

Abstract:A rapid procedure is described for the preparation of extracts suitable for the enzymic-fluorometric analysis of adenine nucleotides and glycolytic and other phosphorylated intermediates, as well as citrate, in small samples taken from mature and senescent tobacco leaves. High recoveries of added adenine nucleotides, citrate, phosphoenolpyruvate, inorganic orthophosphate, and fructose diphosphate indicated that there were insignificant losses during extraction and that interfering substances were removed. The method utilizes extraction with trichloroacetic acid, removal of the latter with ether, and pretreatment of the resulting extract with polyvinylpyrrolidone and DEAE-cellulose. Extensions of the procedure to potato tuber tissue and to the preparation of extracts for chromatography are described.
