枸杞提取液对实验性肾动脉狭窄性高血压损伤鼠的治疗

摘    要:目的:探讨枸杞提取液对高血压疾病的治疗作用和可能的机制.方法:将30只健康SD大鼠随机分为正常组,对照组和治疗组,每组10只.对照组和治疗组建立肾动脉狭窄性高血压模型,治疗组每日给予枸杞提取液灌胃处理,对照组给予生理盐水灌胃处理.饲养一个月后,取SD大鼠心脏、肾脏、大脑做组织病理学方面的检测.结果:与对照组相比较,治疗组血压明显下降(P<0.05),血浆NO合酶cGMP含量明显上升(P<0.05),心肌细胞横截面积有统计性差异(P<0.05);心肌组织结构接近正常,且脑血管病理改变轻.对照组的肾、心、脑组织内cGMP水平与正常组相比显著升高,治疗组cGMP水平较对照组降低,且差别有统计学意义.与对照组和正常组相比,治疗组大鼠心、肾凋亡阳性细胞计数差异均有统计学意义(P<0.05).凋亡因子Bcl-2水平在脑组织内表达显示了组间的差异显著.结论:枸杞提取液对肾源性高血压造成的脏器损害具有保护作用,能够预防由于氧自由基造成的损伤,其作用机制与调节血管活性物质水平和抗氧自由基损伤有关.

Abstract:Objective:To investigate the effect of Lycium Chinensis/Chinese Wolfoerry Extract on Rats with Experimental Renal Artery Stenosis Hypertension,and to explore its possible mechanism.Methods:30 healthy SD rats were randomly divided into three groups:normal group,hypertension group and therapy group(each group has 10 rats).2-clip experimental renal artery stenosis hypertension models were established in the hypertension group and therapy group.The therapy group were treated with Lycium Chinensis/Chinese Wolfberry Extract every day,while hypertension group were treated with the same volume of isotonic Na chloride.A month later,the changes of histopathology and the apoptosis in the kidney,the heart and the brain were observed.The changes of the level of NO,CGMP and SOD in the three kinds of tissue and bcl-2 were also detected by the method of immunochemistry.Results: Compared with the hypertension group,the blood pressure of the therapy group decreased significantly(P<0.05),and the plasma level of CGMP and NO increased(P<0.05).The pathological changes in kidney of the therapy group were fewer than that of the hypertension group,and the average cross section area of the therapy group was smaller than that of the hypertension group(P<0.05).The structure of the heart tissue of the therapy group is near to the normal group.The CGMP level of kidney,heart and brain of the hypertension group were higher than the other groups.The expression of bcl-2 in the brain of the hypertension group was significantly higher than the other two groups(P<0.05).Conclusions:Lycium Chinensis/Chinese Wolfoerry Extract can protect important organs from Renal Artery Stenosis Hypertension and prevent crucial organs from the damage of oxygen flee radicals.The mechanism may be related to the regulating of the active substance levels in blood vessel and the prevention of oxygen free radicals damage.
