温度梯度凝胶电泳技术及应用

摘    要:温度梯度凝胶电泳(TGGE)是一种用于检测核酸序列变异和点突变的电泳方法.该法利用不同构象的核酸分子具有不同的变性温度(Tm)来进行分离.TGGE方法具有分辨能力高、重复性好和节省时间的特点,可广泛应用于分子生物学研究领域.

Abstract:Temperature gradient gel electrophoresis (TGGE) is a new and powerful electrophoresis method for separation of nucleic acids (DNA and RNA) and analysis of sequence variations. The TGGE method uses the melting temperature(Tm) as an important parameter to identify DNA, which differs in sequence among a mixture of molecules of the same size but different conformation. With the advantages of high-resolution capability, high reproducibility and timesaving process, the TGGE technique has been applied broadly in the field of molecular biology.
