鱼藤酮帕金森大鼠呼吸链复合酶Ⅰ、Ⅳ的改变

摘    要:目的研究鱼藤酮帕金森模型大鼠呼吸链复合酶Ⅰ、Ⅳ的变化。方法雄性Wistar大鼠每日颈背部皮下注射鱼藤酮葵花油乳化液2 mg/(kg.d)连续3~5周制备鱼藤酮帕金森模型大鼠;按行为学评分标准记分。模型动物分成高分组、低分组、模型4周组。分光光度法测定呼吸链复合酶Ⅰ、Ⅳ。结果模型低分组肌肉呼吸链复合酶Ⅰ受到明显抑制,停给鱼藤酮4周后肌肉和黑质呼吸链复合酶Ⅰ显著低于正常。而模型高分组肌肉呼吸链复合酶Ⅰ升高,模型各组肌肉呼吸链复合酶Ⅳ均见升高,但黑质未见升高。结论鱼藤酮帕金森模型大鼠肌肉和黑质呼吸链复合酶Ⅰ明显抑制。肌肉见呼吸链复合酶Ⅳ代偿性升高而黑质未见。

Changes of Mitochondrial Respiratory Chain Complex I and Complex IV Activity in Rotenone-Induced Rat Model of Parkinson's Disease

Abstract:Objective To explore the changes of mitochondrial respiratory chain complexes Ⅰ and Ⅳ activity in rats of Parkinson's disease induced by rotenone.Methods The Parkinson's disease model was established by subcutaneous injection of rotenone in oil-emulision(2 mg·kg-1 d-1) to the back of Wistar rats q.d.for 3~5 weeks.The scores of the rats were recorded according to the behavioral score system established in our department.The model rats were divided into the low-score-group,high-score group,and low-score for 4 weeks group.The Activity of complexes Ⅰ and Ⅳ were measured by spectrophotometry.Results The Activity of complex I in the muscle was inhibited in the low-score group and inhibited in the muscle and nigrostriatum at 4 weeks after discontinuation of rotenone.But the activity of complex Ⅰ in the muscle was increased in the high-score group.The activity of complex Ⅳ in the muscle was increased in all groups,but was not increased in the nigrostriatum.Conclusion Those results suggest that the activity of complex Ⅰ in the muscle and nigrostriatum is inhibited in rats with Parkinson's disease induced by rotenone.The Activity of complex Ⅳ is increased in the muscle but not in the nigrostriatum.
