摘    要:本文介绍了日本血吸虫体外培养系统。建立了较适于童虫生长发育的B41培养基。尾蚴经人工方法转变的童虫在体外可发育至雌雄合抱,雌雄生殖器官形成。雌虫可达产卵阶段,但未具备正常的产卵机能。培养的血吸虫在体外至少可存活110天。

Abstract:This paper reports the culture system in vitro of Schislosoma japonicum schistosomula which were transformed from cercariae by syringe method or fresh rabbit serum incubation method.The effects of some nourishment on the growth of schistosomula were compared. RPMI-1640 medium was superior to TC-199 medium for the growth of schistosomula. Rabbit serum was more suitable than new born calf serum. 10% rabbit serum was preferable to 25% or 50% rabbit serum. In addition, insulin, hydrocotisone and serotonin played a role in improving the growth of schistosomula. According to the results, 841 medium was established.The long-term culture of schistosomula produced by mechanical method was observed. The schistosomula were cultured with 841 medium +10% rabbit serum and rabbit blood cells in the incubator at 37C, 5% CO2. The worms began to ingest blood cells on day 5. The intestinal union was seen on day 11 - 12. Pairing of the male and female worms appeared on day 41. The male worms had six to seven testes. -The female worms had one ovary. The female worms began to produce eggs on day 80. Some eggs had a typical lateral spine, but the eggs were abnormal. The schistosomes can be kept alive in vitro for at least 110 days. It showed that the life-cycle stages of S. japonicum which occur in the vertebrate host could almost be completed in vitro.
