摘    要:实验以人胚肺二倍体成纤维细胞(2BS)为材料,分别测定了处于不同代龄及不同生长时期的2BS细胞酸性磷酸酶(ACPase)活性。结果发现随着代龄增高,细胞ACPase活性上升。处于同一代龄的细胞,则接触抑制期细胞的ACPase活性显著高于生长期细胞。接触抑制引起的酶活性增高甚至超过代龄增加而引起的ACPase活性上升。30μg/ml的氯酯醒有抑制细胞ACPase活性的作用。

Abstract:Effects of population doubling level (PDL), living state and centrophenoxine on the ACPase activity were examined in cultured fibroblasts (2BS). The results showed that ACPase activity increased when PDL became higher. In the same PDL group, ACPase ac ivity was much higher in cells of contact inhibition state than that in cells of growth state. Centrophenoxine reduced the ACPase activity in all tested PDLs. It seemed that in addition to the temporary inhibition of cell prolifera tion, the contact inhibition exerted some other effect on the cells, which was more or less like the effects of aging.ACPase as a Iysosomal enzyme may bear some relation to the age pigment, lipofuscin, which is considered to be derived from lysosome. since it is well known that centrophenoxine can inhibit the formation of lipofuscin, it would be interesting to explore whether this drug has an inhibitory effec on the activity of ACPase as well. In our experiment it is evident that centro-henoxine actually tended to inhibit the activity of ACPase in 2BS cells.
