过氧化物酶同工酶应用于甜瓜杂交种真实性与纯度检测研究

摘    要:本研究利用聚丙烯酰胺凝胶电泳法分析了甜瓜(Cucumis melo.L)84—3等六个杂交种及其双亲的苗期过氧化物酶同工酶谱变化,按生长顺序在苗期进行了四次取样,每次取样均取全株。分析结果表明:甜瓜苗期生一片真叶时酶谱丰富,为取样最佳时期,同一组合中,F_1的过氧化物酶同工酶谱与亲本的过氧化物酶同工酶谱是有差异的,存在“杂种酶带”和“互补酶带”,利用过氧化物酶同工酶谱技术对甜瓜84-3等六个组合进行纯度检测结果与田间纯度检测结果相比符合率为99.1%。

Abstract:This study has analysed peroxidase isozyme zymogram changes in seedl-ing of Cucumis melo L. 84-3 etc. 6 hydrids and their parents by polyac-rylamide gel electropnoresis. Samples are take 4 times in seedling accord- ing to their growth stages. Whole plants are taken in each time sampling. Analysis result showed that zymogram is rich rnd it's the best time to sample in the time of one euphylla. In the same group, there are discrepance on the zymogram patterns of F1 generations and its parents, exist "hybrid band" and "complementary band" in the. F1 generationes. Comparing pcroxiclase iso-zyme purity test result with field's result, the corresponding rate is 99.1%.
