C-mye和PTTG在鼻腔内翻乳头状瘤中的表达及临床意义

摘    要:目的探讨原癌基因C-myc和垂体瘤转化基因(pituitarytumortransforminggene,PTTG)在鼻腔内翻乳头状瘤(sinonasalinvertedpapilloma,SNIP)中的表达及临床意义,希望能找到C—myc原癌基因和PTTG与鼻腔内翻乳头状瘤发生发展及预后的关系。方法收集2008~2012年在武汉市中心医院病理科诊断为鼻腔内翻乳头状瘤的存档蜡块20例作为实验组,另取瘤体周围组织5例作为对照组,采用免疫组化SP法检测出C—myc和PTTG的相关抗原。在显微镜下测出每个视野下阳性反应的平均光密度、阳性反应面积和所有细胞的总面积.计算出阳性面积率。采用HPIAS-2000高度清晰度彩色病理图文报告管理系统对C—myc和PTTG在各组中的表达进行定量分析。结果实验组、对照组中C—myc表达的平均光密度为0.3290±0.0218、0.0854±0.0112;实验组、对照组中C-myc表达的阳性面积率为0.2873±0.0152、0.0754±0.0124,两组比较有显著性差异(P〈0.05);试验组、对照组中PTTG表达的平均光密度为0.2850±0.0381、0.0953±0.0127;试验组、对照组中PTTG 表达的阳性面积率为0.3522±0.0365、0.1108±0.0146,经单因素分析两组比较有显著性差异(P〈0.05)。结论C-myc和PTTG的检测尤其联合检测,有助于SNIP的诊断,其对SNIP的预后评估和生物学行为有一定参考意义。

Abstract:Objective To investigate the expression of C-myc oncogene and pituitary tumor transforming gene (FITG) in sinonasal inverted papilloma (SNIP) in an attempt to examine the role of C-myc and PTTG in the occurrence and development of SNIP. Methods Twenty SNIP samples (from 2008 to 2012) were collected from the Department of Pathology of Wuhan Central Hospi- tal. The expression of C-myc and PTrG was immunohistochemically detected. Five samples harvested from the tissues adjacent to the papilloma served as control. The average optical density and the posi- tive area rate of C-myc and FTTG proteins were measured by using image analysis (HPIAS-2000). Results The average optical density of C-myc was 0. 3290±0. 0218 in SNIP group, and 0. 085 4 ±0. 011 2 in control group (P 〈0.05) . The positive area rate of c-myc was 0. 287 3±0. 015 2 in SNIP group, and 0. 075 4±0. 012 4 in control group ( P 〈 0. 05 ). The average optical density of FTTG was 0. 285 ±0. 038 1 in SNIP group and 0. 095 3 ±0. 012 7 in control group (P 〈0. 05). The positive area rate of PTTG was 0. 352 2±0. 036 5 in SNIP group and 0. 1108 ±0. 0146 in con- trol group (P 〈0. 05 ). Conclusions C-myc oncogene and FTTG, especially when used in combi- nation, may become important biomarkers for histopathological diagnosis of SNIP. They may helppredict the biological behaviors and prognosis of SNIP.
