抗肿瘤活性海洋放线菌的筛选及菌株HGF26的初步鉴定

摘    要:对采自连云港海域的海泥样品进行放线菌选择性分离,用其发酵液进行抗肿瘤活性筛选,并对活性较好的菌株HGF26进行了初步鉴定。从海泥样品中共分离得到放线菌78株,以人肝癌细胞HepG2为靶标,活性筛选得到细胞毒活性达60%以上的阳性菌株3株,以其他5株肿瘤细胞为靶标的复筛表明,菌株HGF26的发酵液对多种肿瘤细胞具有显著的细胞毒活性,其中对胃癌细胞BGC823的细胞毒活性为79%,活性产物具有较好的酸碱和热稳定性。通过对菌株的培养特征、形态特征、生理生化特征和细胞壁成分分析,将菌株HGF26初步鉴定为微白黄链霉菌的海洋变种。

Abstract:Actinomycete selective isolation was carried out with marine mud samples collected from sea area in Li-anyungang, a screening for antitumor activity with their fermentation broth, and a better strain in the antitumor activity HGF26 was initially characterized. Altogether 78 strains were isolated from the mud samples, 3 positive of them were screened to have over 60% of cytotoxic activity taking human hepatoma cell HepG2 as a target; it was also indicated by re-screening with another five strains tumor cell that fermentation broth of strain HGF26 had a strong cytoxic activity against many kinds of tumor cell, the cytotoxic activity against gastric carcinoma cell BGC823 was as high as 79% , and the active product having acid-and-alkaline and heat stability, h was characterized that HGF26 as Streptomyces al-bidoflavas var. marinus through culture, morphological, physiological and biochemical features, and cell wall compo-nent analysis.
