Abstract:Tripartite motif-containing protein 15 (TRIM15) is a member of the TRIM family, which is a class of proteins with E3 ubiquitin ligase activity. The function of TRIM15 in tumors is rarely reported. This study is intended to explain the role of TRIM15 in hepatocellular carcinoma. Nuclear and cytoplasmic fractionation and immunofluorescence assays confirmed that TRIM15 was located in the nucleus and cytoplasm. We designed hairpin RNA (shRNA) to knockdown TRIM15 in hepatocarcinoma cell lines. After knocking down TRIM15, cell growth curve and clone formation assays showed that cell proliferation was significantly inhibited (P<0.05). Cell cycle analysis by flow cytometry showed that knockdown of TRIM15 blocked cell cycle in the G0/G1 phase (P<0.05). After knocking down TRIM15, the expression of Cyclin A, CDK2, Rb, P-Rb, and E2F1 were down-regulated, while the expression of CKI p16, p21, and p27 were significantly up-regulated. These findings suggest that knockdown TRIM15 induces cell cycle arrest of liver cancer cells, which indicates that TRIM15 can be used as a feasible target for the treatment of hepatocellular carcinoma.
