HCV E蛋白基因的变异及变异的意义

摘    要:目的分析不同来源HCV的不同亚型(不同型别之间的不同亚型)及同一亚型之间E蛋白基因的变异及变异的意义。方法使用GenBank公开发表的不同来源HCV的不同亚型及同一亚型之间全基因序列为材料,用CLustalw2软件分析序列之间的同源性。结果不同亚型之间E蛋白基因变异达到了70.5%(E1蛋白基因)和59.8%(E2蛋白基因);不同亚型的E2蛋白基因有两个高度变异区,相同的亚型之间只有一个高变区;伺一亚型之间E蛋白基因的变异率为11.8%-16.8%(E1蛋白基因)和18.4~29.9%(E2蛋白基因)。结论根据相同亚型之间E蛋白基因的变异较小的特点,可以利用不同亚型病毒的抗原表位发展多价混合多肽疫苗或多基因混合疫苗,可能会对多种不同亚型病毒的感染起到免疫保护作用。

Abstract:Objective To analyze the variation of E protein gene of HCV from different sub-types and identical hypotypes and the significance of the variance. Method The entire gene sequence of HCV collected from the public database of GenBank was used as the material, and the homology of these sequences was analyzed with the CLustalw2 software. Result The E protein gene variations among different hypotypes were up to 70.5% ( E1 protein gene) and 59.8% ( E2 protein gene) ;E2 protein genes of different hypotypes had two high variation areas ,while that of the same hypotype had only one high variation area;the variation rates of E protein gene from identical hypotypes were 11.8% - 16.8% ( E1 protein gene) and 18.4 - 29.9% ( E2 protein gene). Conclusion Based on the smaller gene variation of the E protein of identical hypotypes, the antigen epitope of hypotype to develop multi-valent polypeptide vaccine or multi-gene mixed vaccine, which could have a protective immune function to infections caused by many kinds of different hypotype viruses.
