Abstract:Recently, it has been successful in the isolation and culture of pluripotential embryonic stem cell lines (i.e. ES cell lines). ES cells are similar in phenotype to embryo cells, and their pattern of differentiation closely resembles that of the normal embryos. Because of this, ES cells provide a unique model system which has a variety of experimental applications, including chromosome aberration, gene expression, development of lethal mutations and transgenic animal production etc. The procedure for the derivation of stem cells from normal fertilized embryos has been extended for parthenogenetically-derived embryos. The establishment of parthenogenetic ES cell line not only confirms that cells from parthenogenetic embryos have the potentialities of development and differentiation, but demonstrates that it is possible to derive cell lines of homozygous diploid genetic constitution from haploid embryos as well. This paper compares the results of several methods of establishment of ES cell colonies from parthenogenetically-derived blastocysts.
