蛋氨酸高产酵母突变株的推理筛选

摘    要:以啤酒酵母(Saccharomycescerevisiae)菌株Ⅰ为出发株,经紫外线处理后,在含可完全抑制出发株生长的乙硫氨酸(浓度为2g/L)的基本培养从上筛出14株生长良好的突变株,其中I-2,I-9株还能在无硫基本培养基上良好生长。经氨基酸自动分析仪检测,I-2株菌体蛋白中的蛋氨酸含量(质量由分比)由I株的1.93增年9.28。

Abstract:The cells of Saccharomyces cerevisiae strain I was incubated on mineral medium (MM)which conained 2. 0g/L of ethionine after treated with ultraviolet. The concentration ofethionine inhibits the growth of the strain. Fourteen well - growing colonies was screenedfrom the basic medium where could totally inhibit the growth of the starting strain. Reason-ably, they are ethionine resistant mutants. Two of the mutants I - 2 and I - 9 also grew wellin MM lacking sulfur, and I - 2 was confirmed as methionine high - yield strain with auto-matic amino acid analyzer to analyze its amino acid to the whole cell protein. The content ofmethionine increased fon 1. 93 (w/w, % ) of strain I to 9. 28 (w/w, % ).
