ATP敏感性钾通道开放剂对脑缺血再灌注损伤的保护作用

摘    要:目的探讨ATP敏感性钾通道开放剂对大鼠局灶性脑缺血再灌注损伤的保护作用。方法40只Wistar雄性大鼠随机分为四组:A组(假手术组)、B组(缺血组)、C组(KATP开放剂治疗组)及D组(KATP开放剂 阻断剂治疗组)。应用线栓法制备大鼠大脑中动脉缺血模型(MCAO),应用TUNEL法检测神经元凋亡,应用免疫组化方法检测Caspase-3蛋白表达,并观察脑梗死体积及神经功能缺损评分。结果(1)C组脑梗死体积显著小于B、D组(P<0.01),B、D组之间无显著性差异(P>0.05);(2)C组神经功能缺损程度较B、D组显著减轻(P<0.05),B、D组之间无显著性差异(P>0.05);(3)C组神经元凋亡数较B、D组显著减少(P<0.01),B、D组之间无显著性差异(P>0.05);(4)C组Caspase-3蛋白表达显著少于B、D组(P<0.01),B、D组之间无显著性差异(P>0.05)。结论KATP通道开放剂能显著减轻脑缺血再灌注后脑梗死体积、改善神经功能缺损程度、减少Caspase-3蛋白表达、抑制神经元凋亡,对脑缺血再灌注损伤发挥保护作用。

Abstract:Objective To study the effects of ATP sensitive potassium channel(KATP) opener on focal cerebral ischemia-reperfusion. Methods 40 male Wistar rats were randomly divided into four groups: A (sham-operated group); B (ischemia-reperfusion group); C (KATP opener treatment group) and D (KATP opener and blocker treatment group). The middle cerebral artery occlusion (MCAO) models were established by using the intraluminal suture occlusion method, the infarct volumes were studied by TTC staining, the scores of neurological function deficits were evaluated, neuronal apoptosis was detected by TUNEL staining, and the expression of caspase-3 protein was detected by immunohistochemical staining. Results (1) The infarct volumes of group C were significantly less than those of groups B and D (P<0.01). There were no differences between B and D (P>0.05). (2) The neurolgical function deficits of C were significantly less than that in B and D (P<0.05), there were no differences between B and D (P>0.05). (3) The number of apoptotic neurons in C was significantly less than that in B and D (P<0.01), with no differences between B and D (P>0.05). (4) The expression of caspase-3 protein in C was significantly less than that in B and D (P<0.01), with no differences between B and D (P>0.05). Conclusion The KATP opener can significantly decrease infarct volume, neurological function deficits, neuronal apoptosis and the expression of caspase-3 protein, which indicates that the KATP opener has a protective effect on cerebral ischemia-reperfusion.
