摘    要:芦笋(石刁柏)栽培品种是较好的蔬菜国外已研究了它的试管苗繁殖技术和单倍体育种。其总氮含量之中,蛋白质氮占四分之三,非蛋白质氮占四分之一。Draper等(1983)曾观察到根癌农杆菌(Agrobateriumtu mefaciens)能附着于从石刁柏叶状枝分离出的细胞之细胞壁。我们设想,芦笋也可能是研究赖氨酸积累或Ti质粒转化单子叶植物的适宜材料。——

Abstract:Asparagus (Asparagus officinalis L. cv.Marry Washington No.500) callus cultures derived from the shoot tips of a female plant were treated by pre- selection in the 1-4-oxalysine (OL, a lysine analog) containing medium, in which the growth of the original type calli were inhibited,for 5 passag- es.The small pieces(<1 mm) of the callus culture were shaked in 1% ethylmethane sulphonate for one and an half hours.The surviving calli were selected out and subcultured suc- cessively on the selecting medium con- taining OL with the concentration in- creased stepwise (2.59 mM,5.18 mM and 10.37 mM) at two or more passages intervals.After the treatment the surviving calli were divided into two portions:one was inoculated to OL (10.37 mM) containing medium and subcultured continuously,denoted line Of2";another was on the medium with- out OL and subcultured successively, denoted line Of2. When the original type f2,line Of2 and line Of2' (the 1st passage of Of2" which was just return back to the medium without OL) were eompared simultaneously on different OL con- centrations,the Of2 which had been subcultured through 6 passages of 24 weeks still maintained resistance to OL. The resistant line Of2 was not cross resistant to ethionine or a-methyl- tryptophan (Table 1)and required 50% growth inhibition concentration of OL which was 10 times and more as that of line f2.(Fig.1) In comparison amino acid analysis,by LKB Model 4101 amino acid analyzer,with the original line f2,the lysine content in line Of2, increased.It seems that in spite of a fair high level of lysine occurred in the original line f2,the accumulation of more lysine could happen in the OL-resistant line Of2 from asparagus (Table 2).
