抗氧赖氨酸的芦笋细胞系(简报)

摘    要:芦笋(石刁柏)栽培品种是较好的蔬菜国外已研究了它的试管苗繁殖技术和单倍体育种。其总氮含量之中,蛋白质氮占四分之三,非蛋白质氮占四分之一。Draper等(1983)曾观察到根癌农杆菌(Agrobateriumtu mefaciens)能附着于从石刁柏叶状枝分离出的细胞之细胞壁。我们设想,芦笋也可能是研究赖氨酸积累或Ti质粒转化单子叶植物的适宜材料。——

Abstract:Asparagus (Asparagus officinalis L.cv.Marry Washington No.500) calluscultures derived from the shoot tips ofa female plant were treated by pre-selection in the 1-4-oxalysine (OL,a lysine analog) containing medium,in which the growth of the originaltype calli were inhibited,for 5 passag-es.The small pieces(<1 mm) of thecallus culture were shaked in 1%ethylmethane sulphonate for one andan half hours.The surviving calliwere selected out and subcultured suc-cessively on the selecting medium con-taining OL with the concentration in-creased stepwise (2.59 mM,5.18 mMand 10.37 mM) at two or more passagesintervals.After the treatment thesurviving calli were divided into twoportions:one was inoculated to OL(10.37 mM) containing medium andsubcultured continuously,denoted lineOf2";another was on the medium with-out OL and subcultured successively,denoted line Of2. When the original type f2,line Of2and line Of2' (the 1st passage of Of2"which was just return back to themedium without OL) were eomparedsimultaneously on different OL con-centrations,the Of2 which had beensubcultured through 6 passages of 24weeks still maintained resistance to OL.The resistant line Of2 was not crossresistant to ethionine or a-methyl-tryptophan (Table 1)and required 50%growth inhibition concentration of OLwhich was 10 times and more as thatof line f2.(Fig.1) In comparison aminoacid analysis,by LKB Model 4101amino acid analyzer,with the originalline f2,the lysine content in line Of2,increased.It seems that in spite ofa fair high level of lysine occurred inthe original line f2,the accumulationof more lysine could happen in theOL-resistant line Of2 from asparagus(Table 2).
