癫痫模型大鼠海马原癌基因c-fos mRNA和蛋白质表达时程的研究

摘    要:为了探讨癫痫发病机理与原癌基因的关系,将马桑内酯注入侧脑室,于大鼠癫痫发作后不同时间点取材海马,用Northern印迹杂交和Western印迹分析技术对海马c-fosmRNA和Fos蛋白表达的动力学进行了定量研究,激光扫描仪定量分析表明:(1)c-fosmRNA在0.5h表达最高,依次渐减,至4h几无表达;(2)Fos蛋白在1h开始表达,2h最多,以后渐少。以上结果表明:c-fosmRNA和Fos蛋白的表达随癫痫的发生发展出现规律性变化,c-fosmRNA是快速,一过性表达,Fos蛋白的表达滞后且延长。提示:c-fos在癫痫发病的基因机制中发挥作用,本文对此进行了讨论。本实验为深入研究癫痫发病的分子机制奠定了基础。

Abstract:In order to investigate the relationship between the mechanism of epileptogenesis and proto-oncogenes, the dynamic alterations of expression of c fos mRNA and Fos protein in hippocampus of rats were quantitatively studied using Northern blot hybridization and Western immunoblot at limited time after the epilepsy attack when Coriaria lacton was injected into lateral cerebroventricle. The quantitative analysis using a laser scanning densitometer showed that the expression of c fos mRNA was maximal at 0 5 h, decreased graduallg and was almost nil at 4 h after the inducer being injected and that the expression of Fos protein began at 1 h, reached maximal at 2 h, then decreased. It is concluded that regular changes of the expression of c fos mRNA and Fos proteim take place with the epileptic start and development, in which the former is rapid, transient and the later is delayed and protracted. The present study suggests that c fos plays an important role in the molecular mechanism of epileptogenesis, which has been disscussed in the present paper.
