分枝杆菌Ag85A DNA疫苗对膀胱癌小鼠细胞免疫功能的影响

摘    要:为探讨分枝杆菌Ag85A DNA疫苗能否提高荷膀胱癌小鼠保护性免疫应答水平,将1×10^6个MBT-2细胞注射于C3H/HeN小鼠的右侧背部皮内,待肿瘤长至直径约5~8 mm时,将动物随机分成实验组、空质粒组和空白对照组3组,分别于小鼠双侧股四头肌内注射Ag85A-V1 Jns.tPA质粒,V1 Jns.tPA质粒总量0.1 mL(100μg),或生理盐水0.1 mL,每14 d 1次,共3次,末次免疫后第14天分别检测T细胞亚群数量、NK细胞活性、FasL mRNA表达情况、淋巴细胞增殖水平及肿瘤重量。结果显示,实验组较空白质粒组和空白对照组免疫指标均无明显增高(P〉0.05),提示肌肉注射Ag85A DNA疫苗不能明显提高荷瘤小鼠免疫功能。

Abstract:In order to investigate whether Ag85 DNA Mycobacterium vaccine could stimulate the protective immune response level in bladder tumor-bearing mice, 1×10~6 MBT-2 cells were injected subcutaneously into right back of C3H/HeN female mouse. When the diameter of tumor reached up to 5~8 mm, the mice were randomly divided into three groups, i. e. experiment group, empty plasmid control group, and blank control group.Subsequently the mice were injected with 100 μg Ag85A-V1Jns.tPA plasmid, V1Jns.tPA plasmid with total amount 0.1 mL (100 μg), or saline into both quadriceps every 14 day intervals, totally three times.14 days after the last vaccination the mice were sacrificed . The quantities of CD4+T cell and CD8+T cell and the cytotoxicity of NK cells and the expression of FasL mRNA and the lymphocyte proliferation of spleen cell as well as the weight of tumor were tested. The results showed that the immune fuction of experiment group had no significantly increased than empty plasmid control group and blank control group. Indicating that intramuscular injection of Ag85 DNA vaccine could not obviouosly stimulate the protective immune response in bladder tumor-bearing mice.
