晶状体生化的研究——亚硒酸钠诱发大鼠白内障的研究

摘    要:我们给出生后10日大鼠皮下注射亚硒酸钠,按隔日每公斤体重3.48mg(20μ moles)诱发大鼠产生白内障获得成功。我们也观察到此种白内障大鼠晶状体中可溶性蛋白质含量下降,经Sephadex G-200柱层析可见β_H、γ晶体蛋白降低,α晶体蛋白相对升高。10%聚丙烯酰胺凝胶电泳及等电聚焦电泳发现γ晶体蛋白改变明显,这些变化和我们先前报告的腹腔注射半乳糖诱发大鼠白内障的结果是一致的。

Abstract:We have successfully induced cataract by every other day subscutaneous injection of sodium selenite at the Ievel at 3.48 mg per kilogram body weight to suckling rats of 10 days after birth. We have observed that the amount of soluble proteins of the cataractous lens was definitely lower than that of normal. Af er analyses of the eluates from columns of sephadex G-200 fine, We have found that the amounts of βH and φ crystallines were decreased and that of α crystalline was relatively increased. v crystalline showed definite change both in 10% polyacrylamide disc gel electrophoresis and gel isoelectric focusing electrophoresis. These results are consistent with those of the galactose induced catarat previously reported from this laboratory.
