Abstract:Methods for the preparation of mitochondria from Prototheca have been investigated and the state of these organelles prepared by various means compared by electron microscopy and measurement of succinoxidase activity. Ubiquinone and cytochrome (a+a3), b and c were present. The mitochondria oxidized NADH and succinate, and mammalian cytochrome c would act as an electron acceptor in the system. The P:O ratios for various tricarboxylic acid cycle intermediates and related metabolites were determined.
