Abstract: | Gene reconstruction analysis, together with the characterization of a collection of genomic clones, have shown that 28 kDa glutelin-2 from maize is coded by a single gene. The similarities in the pattern and relative intensity of bands obtained in genomic blot analysis among endosperm and embryo suggest that no gene amplification or reorganization are associated with the specific expression of the 28 kDa glutelin-2 gene. Endosperm and embryo show clear differences in their DNA methylation pattern when analyzed with the isoschizomeric pair of restriction enzymes MspI and HpaII. Nevertheless, no differences in methylation at CCGG sites have been found in a 0.6 kilo base pair (kb) region upstream the 5′ end of the 28 kDa glutelin-2 gene. |
