Abstract: | The DNA sequences of the spacers between the 5S ribosomal RNA genes were determined for the cereals maize, barley, soghum,
rye, rice, oat, and wheat. Species-specific primers were designed from the spacer region. PCR with these primers and a common
primer from the conserved 5S ribosomal RNA gene sequence was investigated as a method for detection of the seven cereal species.
DNA from these species could be specifically detected in mixtures. This technique could find application in the determination
of the composition of admixtures or processed cereal products. The protocol described has potential for general application
in the identification of plant species. |
