The functions and regulation of tryptophan pyrrolase.

The regulation and functions of rat liver tryptophan pyrrolase are reviewed. The enzyme is regulated by four mechanisms: hormonal induction by glucocorticoids, substrate activation and stabilization by tryptophan, cofactor activation by haem and feedback inhibition by NAD(P)H. Possible functions of the pyrrolase are the detoxication of tryptophan and the regulation of brain 5-hydroxytryptamine synthesis, liver haem metabolism, synthesis of nicotinamide-adenine dinucleotides (phosphates) and hepatic gluconeogenesis.It is suggested that the regulation and functions of tryptophan pyrrolase are physiologically interrelated, and that the enzyme may play important roles in vital body processes.
