共聚焦显微镜检测细胞蛋白分布的研究

摘    要:为使学生对位于细胞不同位置的蛋白的分布情况有直观的理解,针对本科细胞生物学基础实验开设的共聚焦显微镜课程内容作了改进研究:将绿色荧光蛋白(GFP)表达质粒转染入细胞(相当于胞内蛋白);用免疫荧光方法标记细胞膜蛋白;FITC偶联的鬼笔环肽标记的细胞骨架蛋白,用DAPI染核,然后利用共聚焦的特有功能对细胞进行局部放大及分层扫描。实验结果得到了较为清晰的图像。

Abstract:To understand the location of proteins in the cell better,the experiment about the using confocal microscope was improved.Green fluorescent protein(GFP) expression plasmid was transfected into cells(the equivalent of cellular protein);membrane proteins were stained by immunofluorescence method,cytoskeletal proteins were labeled FITC-coupled philloidin.The cells were stained with DAPI,and then scanned by confocal microscope.The unique capabilities of the confocal,cell layer scanning and local zoom,were used....
