APP/PS1转基因小鼠海马结构UCH-L1的表达及亚甲蓝对其改变的影响

摘    要:目的探讨亚甲蓝(MB)对APP/PS1转基因小鼠记忆相关蛋白泛素羧基末端水解酶1(carboxyl-terminalhydrolase-L1,UCH-L1)在海马结构的表达及记忆改善的影响。方法 3月龄APP/PS1转基因小鼠及相同品系的野生小鼠分为3组,每组10只:治疗组,APP/PS1小鼠口服亚甲蓝(25mg/kg/d)4个月;模型组,APP/PS1小鼠无药物干预;对照组为正常野生小鼠。待三组小鼠均为7月龄时,跳台实验测试三组小鼠的学习记忆能力;Western blot及免疫荧光技术检测海马结构UCH-L1的含量变化。结果亚甲蓝可以减少小鼠跳台试验错误次数,延长小鼠跳台试验的潜伏期(P<0.01)。亚甲蓝治疗组海马结构的可溶性UCH-L1含量明显增多(P<0.01)。结论亚甲蓝可能是通过上调海马结构可溶性UCH-L1的表达改善APP/PS1小鼠的学习记忆能力。

Abstract:Objective To investigate the effects of methylene blue (MB) on the expression of UCH- L1 in the hippocampal formation of APP/PS1 transgenic mice. Methods APP/PS1 mice and wild type (WT) mice were divided into 3 groups. MB group (APP/PS1 mice treated with MB 25mg/kg/d,P. O. ), model group (APP/PS1 mice without MB treatment), and control group (WT mice). The ability of learning and memory was assessed by the step down avoidance test. The expression of UCH-L1 was measured by Western blotting and immunofluorescence. Results After MB treatment, the number of error was decreased and latency was prolonged in the step down avoidance test (P〈0.01). Compared with that of the model group, the expression of soluble UCH-L1 in the MB group increased significantly. Conclusion MB improves the ability of learning and memory by up-regulating the expression of soluble UCH-L1 in the hippocampal formation.
