A rapid assay for pectinesterase activity which can be used as a prescreen for pectinesterase inhibitors.

A rapid assay for inhibition of pectinesterase has been developed to facilitate large scale screening for possible inhibitors. The assay gives quantitative results which can be confirmed by other assay techniques. The assay uses an agar plate in which pectin and an indicator have been incorporated. The hydrolysis of pectin to pectinic acid by pectinesterase causes a lowering of the pH of the medium and produces a color change. The area of the color change produced by the hydrolysis of pectin is directly proportional to the activity of the pectinesterase.
