| 摘 要： | 本文报道了一种较简便的从猪胸腺中分离纯化末端脱氧核苷酰转移酶(TdT)的方法。经一次磷酸纤维素柱层析,使TdT与DNA聚合酶分离;经三次柱层析,可获得SDS-电泳纯,分子量约60K的产品。其酶学性质与牛胸腺TdT相似。

| Abstract: | This is a report on the purification of terminal deoxynucleotidyl transferase (TdT) from pig thymus. A simplified procedure for purification of TdT is established. In this procedure DNA polymerase and TdT in cell extracts have been deparated by P-11 cellulose column. After three chromatographic steps, the TdT ace tivity in the last fraction has increased 13,000-fold as compared with the crude-extract. The specific activity of the purified pig TdT is about 613 units/mg of protein. The purified pig TdT seems to be homogeneous, as shown by SDS-gel electrophoresis, and contains a single polypeptide about 60K in molecular weight. The enzymatic properties of pig TdT are very similar to those of the enzyme in calf. |
