高等担子菌双重营养缺陷突变菌株的分离

摘    要:带有单一营养缺陷的凤尾菇和裂褶菌的单核体菌株经亲和性交配,各自交配产生后代,从中分离出遗传特性稳定,生理特征表型正常的双重缺陷营养害变型菌株,为原生质体融合育种研究提供了可靠的亲本菌株。

Abstract:In culer to get double auxotrophic muntants, two compatible auxotrophic mutants of (Pleurotus sajor-caju were mared with each other on CMA plates and also were done for Schizophyllum commmune. The fruit bodies were induced from those matings and the spores were collected from those produced fruit bodies, and then the monokaryons were isolated from those spores on CMA. Those monokaryons were subcultured onto the MMA, MMA one nutrient which was deficient for one parent and MMA another nutrient which was deficient for the other parent The monokareons that could not grow on these three kinds of media were suposed to be double mutants and this was further confirmed by subculturing them onto MMA supplemented by the two nutrients which were deficient for the two parents. A rather number of stable double auxotrophic mutants were selected and they are useful as a start strain in protoplast fusion study.
