饥饿及复投喂对大黄鱼肌肉生长抑制素1型和2型基因表达的影响

摘    要:采用荧光实时定量PCR技术研究饥饿及复投喂对大黄鱼不同组织中两型肌肉生长抑制素(Myostatin, MSTN)基因表达的影响。结果显示, 肌肉中MSTN-1和MSTN-2在饥饿035d期间, 表达量先升高后降低, 分别在21d和28d达到最高, 复投喂时期逐渐下降; 脑中两型MSTN表达变化不同, 但都在饥饿35d表达量最低, 复投喂时期表达显著上升, 与对照组有显著差异; 脾脏中MSTN-1在饥饿28d表达量最高, MSTN-2在饥饿21d表达量最高, 复投喂时期表达都逐渐降低; 肾脏中两型MSTN表达变化相似, 都在饥饿21d降到最低, 然后升高; 肠中1型和2型分别在饥饿14d和7d表达量最低, 在28d表达量都达到最高; 肝脏中两型MSTN都是在饥饿21d表达量最低, 之后显著升高, 复投喂时期表达上升。结果提示两型MSTN在不同组织中的功能可能存在差异。

Abstract:In this study, we used real-time PCR to investigate the effects of fasting and re-feeding on the expression of two MSTN genes in large yellow croaker (Larimichthys crocea). The results showed that during the 35-day fasting treatment, the expression of MSTN-1 and MSTN-2 in skeletal muscles firstly increased and then decreased, and their expressions perked on the 21st and the 28th day respectively. The expressions were down-regulated during the re-feeding period. The expression patterns of the two MSTNs were different in the brain, although they both reached the lowest level on the 35th day and then increased after re-feeding. In the spleen the expressions of MSTN-1 and MSTN-2 reached to the highest levels on the 28th and the 21st days respectively, and both declined after the re-feeding. In kidney the expression patterns of the two genes were similar, and they both reached the lowest on the 21st day after fasting followed by an increase. The expression of MSTN-1 and MSTN-2 in the intestine decreased significantly on the 14th and the 7th day respectively, and then peaked on the 28th day. In the liver, the expression of MSTN-1 and MSTN-2 decreased sharply on the 21st day and then increased. The expressions of both genes were up-regulated after re-feeding. Our findings suggested that the functions of the two MSTN genes might be various in different tissues.
