洋葱叶肉原生质体培养再生小植株

摘    要:用纤维素酶(EA 3-867)和离析酶(Macerozyme R-10)酶解洋葱叶肉细胞,游离获得大量具有活力的原生质体。在MS培养基上(附加2,4-D 2,6-BA 0.5 mg/l,原生质体能再生细胞壁,生长,分裂,形成类似球形的愈伤组织;将它们分别移入分化培养基MS_1,MS_2,MS_3,得到再生的小植株。

Abstract:Large amount of viable mesophyllprotoplasts were isolated enzymaticallyfrom onion(Allium cepa L.).The pro-toplasts grew and divided when culturedin thin liquid layers of MS mediumwith 2,4-D 2,6 BA 0.5 mg/1,mannitol0.5 M,sucrose 2%.Calli were subcultu-red on MS medium without mannitol.After transferring to modified MS me-dium(MS_1,MS_2,MS_3)calli regeneratedbuds or bulblets and regenerated plant-lets had been obtained.
