Abstract: | A high-performance liquid chromatographic method has been developed for the resoiusion of DNA bases and deoxyribonucleosides in mixture. The isocratic separation based on ion exelusion was achieved in column of 250-mm length and 2 mm i.d. packed with the spherical, totally porous anex OSTION LGAT 0800 of particle size 10–12 μm. The mobile phase consists of 5 mm ammonium formate pH 4.5 and complete resolution of an eight-component system is possible at a flow rate of 2 ml/h in 100 min. Rapid separation of the components, with exception of Cyt-dCyd, is reached at flow rate of 20 ml/h in 10 min. |
