Assay of free tryptophan in urine by high-voltage electrophoresis

Abstract:A method for the assay of free tryptophan in urine by means of high-voltage paper electrophoresis gives reproducible results comparable to the microbiological test, the only procedure so far found applicable to urine. It was found that a number of substances related to tryptophan may seriously interfere with the microbiological assay. In the technique proposed, the interference of indolic compounds and of urea is obviated, as they are sufficiently separated by the strong electric field and give different colors in the chromatic reaction with PDAB. Tryptophan gives a distinct easily separable spot that can be quantitated by densitometry. Comparison of this method with the microbiological assay of urines from phenylketonurics showed that some nontryptophan materials that appeared on microbiological assay like tryptophan were excreted up to seven times as much as tryptophan. It is likely that one of these is indolelactic acid, but this cannot account for the bulk of the excreted “tryptophan” in phenylketonuria.
