| Abstract: | In this paper, the J6R ( F12 ) promoter, which promotes the expression of gene coding a 147 kilodalton unit of vaccinia virus RNA polymerase, had been cloned from the Tian Tan strain of vaccinia virus with DNA polymerase chain reaction ( PCR ) . This showed that PCR technique can be applied to clone and modify the promoter of vaccinia virus. The functional study of F2 promoter in eukaryocyte indicated that F12 promoter was an early promoter with partial late activity. The activity of J6R promoter that we cloned was lower than that of WR strain J6R promoter reported by Barbeyron. The possible reason of this result and suggestions for further research is discussed |
