Abstract:The molecular weights of rabbit and human hemopexin were investigated by sedimentation equilibrium in the analytical ultracentrifuge. The apparent isopotential specific volume (φ′) of rabbit hemopexin was found to be 0.702 ± 0.003 ml/g which is very similar to the partial specific volume of 0.705 ml/g calculated from the amino acid and carbohydrate composition of both rabbit and human hemopexin. With φ′ = 0.702 ± 0.003 ml/g, the molecular weight of rabbit and human hemopexin is 57,000 ± 3,000. The extinction coefficients (E1cm1%) for rabbit hemopexin containing 1 mole of heme per mole of protein are 21.8 at 280 nm and 19.2 at 414 nm; the apoprotein has an E1cm1% of 19.7 at 280 nm.
