太空诱变哈密瓜两性花性状连锁标记的RAPD分析

摘    要:以哈密瓜品种'早皇后'、太空诱变后两性花株突变体及其后代分离群体为试材,采用BSA方法对哈密瓜两性花性状进行了RAPD分析.研究结果表明:在所筛选的490条10-mer随机引物中,只有S1254在两性花株基因池中扩增到750 bp的多态性条带,而在其它基因型的群体中未扩增到此条带.通过对分离群体及其姐妹系进行单株验证,均获得相同的扩增结果,说明S1254750与两性花性状连锁,该标记与两性花性状的遗传距离为4.5 cM.

Abstract:In the present study,Hami melon 'Zaohuanghou',hermaphrodite mutant after space mutation and its offspring group were investigated.Bulked segregation analysis (BSA) was employed to identify randomly amplified polymorphic DNA (RAPD) markers linked to the hermaphrodite type in Hami melon.The results of the study revealed that a 750 bp polymorphic band was amplified with primer S1254 in the hermaphrodite pool in a total of 490 primers screened,while this polymorphic band were not amplified in other genotype groups.The result was confirmed by the validation of the separation groups and sister groups,RAPD marker S1254750 was proved to be linked to hermaphrodite type,the genetic distance is 4.5 cM.
