我国一株鹦鹉病毒的鉴定及其生化特性

A strain of new virus was isolated fromfledgling budgerigars by authors in 1995. Cytopathic effect became visible after theisolate was passaged 4 generations on budgerigar embryo fibroblast and 5 generations onChicken embryo fibroblast. The SDS-PAGE proved that viral capsid was composed of eightpolypeptides with molecular weight from 60 000 to 14 500 D. The protein was rich in Asp,Glu and Leu, but comparatively poorer in Arg and Met. The ratio of acidic amino acid tobasic amino acid was 1∶2.51. The nucleic acid was dsDNA and existed as supercoiledcircular, relaxed circular and linear molecules. G+C of viral DNA was 45%. The molecularweight of DNA was 3.3×10⁶D by restriction endonuclease analysis. According tostandards of virus classification, it is proved that the isolate was budgerigar fledglingdisease virus which had placed into the Polyomavirus genus of Papovaviridae.
