Abstract: | Summary The ERG10 gene was amplified inS. cerevisiae and inS. uvarum, in order to study the regulation of ergosterol biosynthesis. Transformants with 10 or 20 fold increase in acetoacetyl CoA thiolase activity show the same ergosterol (aerobiosis) and squalene (anaerobiosis) contents as the wild type strains, confirming the presence of limiting steps in the conversion of acetoacetyl CoA into sterols. Measurement of mevalonate and phosphomevalonate synthesis shows that mevalonate kinase could be repressed in transformed strains. |
