Abstract: | α-Amanitin, a potent inhibitor of RNA polymerase II, is found inert against transformed fibroblasts in tissue culture. However, when α-amanitin is synergistically used with amphotericin B, RNA and protein synthesis are strongly blocked. Our data suggest that messenger RNA formation is preferentially inhibited since (1) the total inhibition by α-amanitin was greatly magnified when rRNA synthesis was first blocked with 0.03 μg/ml actinomycin D; (2) mRNA in polysomes was greatly reduced and the size of polysomes diminished after cells were exposed to 2 μg/ml α-amanitin plus 20 μg/ml amphotericin B for 5 h. |
