烟草内源铁蛋白NtFer1和NtFer2的相互作用

摘    要:以2个烟草铁蛋白基因全长序列(NtFer1和Mnr2,GenBank登录号:AY083924和AY141105)为基础,利用细菌双杂交系统分析不同烟草铁蛋白亚基之间及相同铁蛋白亚基之间的互作关系,并利用Northern杂交分析2个铁蛋白基因的特异表达。结果表明,铁蛋白基因NtFer1和Mnr2在叶片中均有表达,同时2种亚基之间存在很强的互作关系,说明在叶片中组成铁蛋白的24个亚基可能有3种类型,或来自单一的NtFer1亚基,或来自单一的NtFer2亚基,也可能来源于不同的铁蛋白亚基。在烟草根部组织中只有铁蛋白NtFer1基因大量表达,而MFPr2基因的表达非常微弱,所以根部的铁蛋白大分子可能由单一的铁蛋白NtFer1亚基聚合而成的。

Abstract:Based on the full length sequence of two tobacco ferritin genes (NtFerl and NtFer2), the differential expression of ferritin genes was detected by Northern blotting, and the protein interaction among the ferritin subunits were analyzed by BacterioMatch two-hybrid assay system. Results indicated that the NtFerl and NtFer2 gene could be higher expressed in leave, and a strong interaction between NtFerl and NtFer2 subunits were detected, and suggested that composed of 24 subunits of ferritin may contain three assemble types: sole kind NtFerl subunit, or sole kind NtFer2 subunit, or multi kind subunits. In the root, only NtFerl gene was expressed, which suggested that composed of 24 subunits of ferritin in root was sole kind of NtFerl subunit.
