膀胱癌H-ras基因点突变和ras癌基因产物P21的表达

摘    要:应用聚合酶链反应(PCR)技术结合地高辛化寡核甘酸探针杂交以及免疫组织化学方法,检测人膀眈癌组织中H-ras癌基因第12位密码子点突变和基因产物P21蛋白的表达.58例膀胱癌组织中有23例出现H-ras基因的点突变,36例P21蛋白表达阳性,5例正常膀胱组织未见点突变和P21蛋白阳性表达.本组膀胱癌第12位密码子点突变和P21阳性表达水平呈正相关,多发生在肿瘤临床T2-T4期和分化差的肿瘤,并且与膀胱癌的预后有关、提示H-ras癌基因点突变和P21蛋白阳性表达是膀胱癌的晚期表现,并在膀胱癌的发生,发展及预后中起着重要作用.

Abstract:We detected mutation at codon 12 of H-ras gene and expression of P21 product of human bladder cancer using polymerase chain reaction (PCR) in combination with oligonucleotide probe hybridization and immunohistochemistry. The mutations were found in 23 out of 58 bladder cancers. P21 positive reactions were showed in 36 out of 58 however in tumors, the 5 normal tissues analyzed, none contain mutation and all were negative for P21 expression. The mutation incidences were positively related to P21 overexpression of bladder cancer, and most of them were of T3-T4 stage and G3 grade, which were assicated with the prognosis of the tumor. Our results suggest that H-ras mutation and P21 positive expression may be a late presentation, both of them play an important role in the development and prognosis of human bladder cancer.
